1) Read a good book on chromatography and/or method validation
2) Choose a suitable detector and column
3) If your compound is ionizable, choose a suitable buffer
4) Start with a generic gradient and see where your compound elutes
5) Refine the gradient
6) If possible convert to an isocratic run
7) Validate the method
